M.Sc. Research Project                                            Guided by Mr.V.P.Khandwekar Title: Comparative study of normal and farmer chromosome banding pattern.

In the present study White blood cells of Normal and Farmer were used for karyotyping. White blood cells are used most frequently because they are easily induced to divide and grow in tissue culture. Here used LSM1077 for centrifugation, heparinized blood, RPMI-1640 media, colcemid solution, hypotonic 0.075 KCL and Co2 incubator for cell culture. Giemsa stain was used for staining. From karyogram analysis, observation is derived that in comparison of both banding patterns of normal as well as farmer. Farmer shows more light bands (euchromatin) as compare dark band (heterochromatin) especially in chromosome no.2 so it concluded that the more gene will be active state of transcription as compared to normal which have less light bands. Further according to relative position of centromere, normal and farmer karyotypes was found to be stable.
